Alnylam Pharmaceuticals [ALNY] — Trade Construction & Risk
2026-07-29 · framework: Criteria, 2026-07-29
This document issues no position verdict. It states what a position would have to look like if a book
chose to take one, what would invalidate the analysis, and what the actual tradable instruments are. The
capital decision belongs to portfolio-book, not here.
1. Vehicle — the chain was pulled, not assumed
criteria.md, Liquidity Criteria: "Any proposed options structure requires the actual chain pulled first —
open interest and quoted size for the specific strikes and expiry." Pulled from Alpaca on 2026-07-29.
Common stock — viable
| 63-day median volume | 1,128,486 shares |
| 63-day median dollar volume | $328.9m |
| A $10m position | 3.0% of one day's volume |
| A $25m position | 7.6% of one day's volume |
| 20-day realised volatility | 42% |
Exit in a stressed tape at 20% of ADV is one to two days for $10m. PASS.
Options — not viable at institutional size, and this is the finding
January 2027 calls (166 days), spot $288.50:
| Strike | Open interest | Bid | Ask | Bid size | Ask size | Spread as % of mid | IV | Delta |
|---|---|---|---|---|---|---|---|---|
| 260 | 5 | 56.17 | 59.64 | 1 | 4 | 6.0% | 53.4% | 0.70 |
| 280 | 3,578 | 45.02 | 49.97 | 17 | 4 | 10.4% | 53.1% | 0.62 |
| 290 | 56 | 40.23 | 44.53 | 3 | 5 | 10.2% | 52.4% | 0.58 |
| 300 | 202 | 35.58 | 39.42 | 5 | 3 | 10.2% | 51.4% | 0.55 |
| 340 | 364 | 21.21 | 24.72 | 3 | 4 | 15.3% | 49.8% | 0.40 |
| 370 | 1,736 | 14.33 | 17.52 | 3 | 3 | 20.0% | 49.6% | 0.30 |
| 400 | 92 | 8.25 | 11.94 | 5 | 4 | 36.5% | 48.0% | 0.22 |
March 2027 calls: total open interest across the entire $260–400 range is 326 contracts, maximum 116 at any single strike.
Assessment. Open interest exists at two January strikes, but quoted size is 1 to 17 contracts and the spread on the most-liquid strike is 10.4% of mid. A 280/370 January call spread — the structure the delta profile would suggest — requires crossing roughly 30% of mid in round-trip friction. Any structure above approximately 50 contracts (~$225k of notional delta) cannot be filled at quoted size. This is the HCA failure mode (maximum 18 contracts across an entire chain) in a milder form, and the conclusion is the same: a vehicle that cannot be filled is not a vehicle.
Implied volatility is 48–53% against 42% realised. Long premium is being sold expensive here. That is a second, independent reason the options route is unattractive even at a size it could absorb.
If a position is taken, the vehicle is the common stock. No options structure is recommended and none is appended to
trade_recommendations.jsonl.
2. Sizing inputs (the book computes the size; these are the inputs)
| Input | Value |
|---|---|
| 20-day realised volatility | 42% |
| Scanner volatility (screen) | 38% |
| Volatility tier | High — inverse-volatility sizing is the active protection per criteria.md |
| Beta | not regressed — no figure asserted |
| Correlation to the existing book | not computed here; portfolio-book owns it |
| Sub-sector for concentration | Pharma (rare disease / RNAi). Neighbours: VRTX, REGN, NBIX, BMRN |
criteria.md on the Downside Criteria: "Inverse-volatility sizing is the active protection, and it works
because a fat-left-tail name is almost always a high-volatility name and is sized down automatically." ALNY
at 42% realised is sized down automatically relative to a 20% name. That is the intended mechanism and no
separate cap is asserted.
3. Entry timing — what Momentum Criteria says, and what it does not
Momentum is MEASURED. It governs when, never whether.
| Metric | Value |
|---|---|
| 12-1 momentum | −9.1% (scanner −9.7%) |
| 12-1 cross-sectional percentile | 38th |
| 6-1 cross-sectional percentile | 29.4th |
| % of 52-week high | 58.7% |
| Above 200-day moving average | No |
| RSI-14 | 51.3 |
| Momentum quintile | 2 |
Read. The name is 41.3% below its 52-week high, in the second momentum quintile, below its 200-day, with RSI at neutral. Revenue is guided +49% and the EV/sales multiple sits at the 2nd percentile of its own 4.5-year range. That is a de-rated grower, not a broken one — the multiple has compressed by roughly 60% from its 2022–2023 norm while the revenue base has quadrupled.
Timing implication, not a veto. A book that scales in rather than taking a full position at once is acting consistently with a 38th-percentile momentum reading. The natural scaling checkpoint is dated: Q2-2026 results, expected late July / early August 2026 (see the calendar — the date is not yet announced and is flagged as expected, not confirmed).
Any language in which momentum blocks or vetoes this name is deleted. Under the retired numbering this
was Gate 6, and its demotion to MEASURED is the type-discipline test case in criteria.md.
4. Invalidation — dated, falsifiable, and specific
These are the conditions under which the analysis in this memo is wrong, not the conditions under which the price falls.
| # | Invalidation condition | How it is observed | When |
|---|---|---|---|
| 1 | FY2026 total net product revenue lands materially below the guided $4,900–5,300m. The entire 12-month target and the demonstrated-CAGR base rest on this. A print below ~$4.4bn breaks both ends. | Q2, Q3, Q4-2026 earnings; guidance revision | quarterly through Feb-2027 |
| 2 | A vutrisiran post-marketing safety signal or an ATTR-CM label restriction. This is the named permanent-loss cause. | FDA communication, label change, 8-K | any time |
| 3 | Total TTR quarterly revenue declines sequentially for two consecutive quarters while ONPATTRO is already at run-off. That would mean AMVUTTRA has hit its diagnosed-pool ceiling far earlier than the mechanism implies. | quarterly revenue summary in the 8-K Ex-99.1 | quarterly |
| 4 | COGS exceeds ~25% of net product revenue without a stated offset. It moved 15.0% → 20.0% in one year on a rising blended royalty rate. At 25%+ the terminal-margin assumption of 35% is no longer reachable and the implied-path test has to be re-run. | quarterly income statement | quarterly |
| 5 | Distributor A concentration rises further, or a channel-inventory correction is disclosed. At 45% of gross revenue a single counterparty now moves reported revenue independently of demand. | 10-K concentration note (annual); DSO in quarterly balance sheet | Feb-2027 / quarterly |
| 6 | FY2027 collaboration + royalty revenue comes in materially above $500m without a new signed agreement. That would mean the lumpy line is being used to bridge a product-revenue shortfall, and the recurring decomposition in this memo would need redoing. | quarterly revenue summary | quarterly |
What is explicitly not an invalidation. A drawdown. ALNY has 42% realised volatility; a 25% decline is roughly a one-standard-deviation half-year move and carries no information about the mechanism. The Downside Criteria is about permanent impairment, not volatility.
5. Downside Criteria — logged for the ledger
Per criteria.md, the bear case, its probability and its named cause are logged on every name so that
ledger_scorer.py can Brier-score them.
| Field | Value |
|---|---|
| Bear case | A vutrisiran post-marketing safety signal or an ATTR-CM label restriction removes the mechanism rather than repricing it. AMVUTTRA's $2.3bn run-rate is impaired and the pipeline is discounted with it. |
| Named cause | Regulatory/scientific event on vutrisiran in ATTR-CM |
| Probability | 10% |
| Drawdown if it occurs | −55% to −65%, to roughly $100–130 (≈4–5x recurring revenue, the trough multiple for a broken single-franchise biopharma) |
| Secondary bear (economic, not permanent) | Three-way branded ATTR-CM price competition (tafamidis, acoramidis) compresses US net price 20–25% from 2028 and caps the TTR franchise near $6bn. Probability 30%. At 8.5x sales — the bottom of ALNY's entire observed range — EV is still ≈$55bn, above today. This is a "dead money" case, not an impairment case. |
| Tertiary | Distributor A (45% of gross revenue) disruption or channel correction. Probability 10%, drawdown −15% to −25%, recoverable. |
| Going concern | No. $3.0bn cash and investments, GAAP profitable, $524.1m FY2025 operating cash flow, $465.4m free cash flow. Not arguable and not argued. |
Honest note on the bear case, since criteria.md records the principal's objection that bear cases are
often used "without much backbone or rigorousness … to exclude good investments": the economic bear case
here does not produce a loss at this price. I have said so rather than manufacturing one. The only case
that impairs capital permanently is a scientific event, and I have given it a probability rather than a
paragraph.
6. Position construction, if a book takes one
Stated conditionally. The memo does not decide this.
| Element | Specification |
|---|---|
| Direction of the research | Positive — Quality PASS, Valuation PASS with a +41.1pp margin, no short mechanism |
| Vehicle | Common stock only. Options are uninvestable at size (§1) |
| Entry | Scale, consistent with a 38th-percentile momentum reading. A first tranche at market; the natural checkpoint for the balance is Q2-2026 results |
| Sizing | Inverse-volatility on 42% realised. High-volatility tier. portfolio-book computes the number |
| 12-month target | $481 (+66.7%); bear $390 (+35.2%); bull $684 (+137.1%) |
| Invalidation level (price) | None asserted as a stop. The invalidations in §4 are evidentiary, not price-based, because a 42%-vol name generates price-based stops as noise |
| Hard evidentiary invalidation | FY2026 product revenue below ~$4.4bn, or an ATTR-CM safety/label event |
| Ledger entry | No options recommendation is appended to trade_recommendations.jsonl. That file is a ledger of specific options trade recommendations and there is none to log. The bear case above is logged for scoring |
7. What a book should watch that this memo cannot resolve
- Consensus. No Street numbers were obtainable (Alpha Vantage quota exhausted by parallel agents). The house-versus-Street question — the one the Consensus Criteria exists to answer — is INDETERMINATE. If the Street is already at $5.1bn for FY2026 and $7.0bn for FY2027, the 12-month target here is a consensus target and not a variant view. That is unknown and is not guessed at.
- The competitive read on acoramidis. ALNY's own corpus is silent (zero mentions in fourteen quarters). BridgeBio's own disclosures are the place to resolve this and were outside this run's scope.
- The blended royalty rate payable on AMVUTTRA. Disclosed to be rising; the rate itself is not given. This is the single largest uncertainty in the terminal-margin assumption.
- Correlation with the existing book. Not computed here. ALNY's correlation with any existing VRTX,
REGN, NBIX or BMRN exposure is a concentration question
portfolio-bookowns.
